PE-22-28 is described as a potent, selective antagonist of TREK-1 (KCNK2) two-pore potassium channels. TREK-1 normally contributes to neuronal “leak” currents that dampen excitability; blocking TREK-1 depolarizes neurons, increases excitability, and is described as increasing firing of serotonergic neurons with downstream elevation of monoaminergic neurotransmission. In preclinical models referenced by the protocol, these changes are associated with rapid antidepressant-like effects, including increased hippocampal neurogenesis and markers of synaptogenesis within days—faster than typical timelines reported for conventional antidepressants.
