PE-22-28 is described as a potent, selective antagonist of TREK-1 (KCNK2) two-pore potassium channels. TREK-1 normally contributes to neuronal “leak” currents that dampen excitability; blocking TREK-1 depolarizes neurons, increases excitability, and is described as increasing firing of serotonergic neurons with downstream elevation of monoaminergic neurotransmission. In preclinical models referenced by the protocol, these changes are associated with rapid antidepressant-like effects, including increased hippocampal neurogenesis and markers of synaptogenesis within days—faster than typical timelines reported for conventional antidepressants. The page also notes activation of CaMKII/CREB signaling, pathways associated with neuronal survival and plasticity. PE-22-28 is positioned as an optimized, shortened analog of spadin with higher potency and longer duration of action. Additionally, the protocol describes neuroprotective findings in stroke models using biphasic dosing concepts.
Minor injection-site reactions (slight redness/tenderness)
No human safety/pharmacokinetic data (research-only evidence base)
- Use aseptic technique: wipe vial stopper with alcohol; use new sterile syringe/needle
- Add diluent slowly down the vial wall to minimize foaming
- Gently swirl/roll until fully dissolved (do not shake)
- Label vial with reconstitution date and concentration; protect from light
- Refrigerate after reconstitution (commonly 2–8 °C) unless protocol states otherwise
- Avoid repeated freeze–thaw cycles
- Bacteriostatic Water for Injection contains benzyl alcohol preservative (multi-dose); follow protocol for beyond-use (many peptide protocols use ~28 days after mixing)
- Avoid benzyl-alcohol-containing diluents in neonates/infants (safety warning for benzyl alcohol)
